Establishing impurity specifications for linker-payloads in antibody-drug conjugates (ADCs) poses significant challenges, especially during the early development stages. At WuXi XDC, we confront these complexities head-on by leveraging specialized analytical methods to evaluate the conjugatable nature of impurities. Our fit-for-purpose strategy utilizes liquid chromatography-mass spectrometry (LCMS) for preliminary identification of reaction impurities, while mock-conjugation experiments help verify their characteristics. This poster will present our innovative approaches to resolving the intricacies of chiral centers, polymeric homologues, and epimerization, ultimately enhancing the quality and consistency of ADC drug substances and products.
