Immunohistology

Microscopic Immunohistochemistry (IHC) and Immunofluorescence (IF) Services

Immunohistology

Histology studies often encounter challenges in achieving rapid, precise evaluation of drug-related toxicity from tissue analysis. At WuXi Biologics, our precise immunohistochemistry and immunofluorescence services ensure timely access to biological samples with insightful data interpretation. We specialize in microscopic analysis and tissue diagnostics to support drug development, offering immunohistochemical analysis for target validation in tumor model design, diagnosis of drug-related toxicity, and pathological assessments to verify both the mechanism of action (MOA) and toxicity of antibody leads. By combining responsive sample management with expert scientific analysis, we generate reliable, high-quality data to support rational study design and accelerate preclinical studies.

Sample requirements:

1.     Tissues must be collected and fixed with 4% PFA for FFPE or frozen in nitrogen for cryosectioning immediately after sacrifice.

2.     For unexpected deaths, issues/organs must be collected promptly to avoid autolysis.

Deliverables:

1.     Histology: Slides and raw staining images, including pathological diagnosis of tissue morphology and immune cell infiltration assessment.

2.     IHC or IF: Slides and raw staining images with quantifiable analysis for antibody distribution and target validation.

Case Study #1: Comprehensive Immunohistochemistry (IHC) and Immunofluorescence (IF) Studies for Tumor Model Selection and Lead Characterization

This case study showcases the application of IHC and IF techniques for tumor model characterization, drug distribution analysis, and immune profiling.

Figure 1: (A) IHC staining was performed to characterize and select tumor models based on antigen expression. (B) Human Fc distribution was analyzed using IHC staining to determine antibody-drug conjugate (ADC) localization within tumor tissues. (C) IHC staining for human CD4 T-cells was conducted to evaluate immune cell infiltration in the tumor microenvironment. (D) Multiplex immunofluorescence was utilized to profile the immune landscape, including DAPI, human CD4, and human CD8 staining.

Case Study #2: Comprehensive Immunohistochemical Staining Capabilities for Drug-Related Toxicity and Tissue Pathology

In this case study, we utilized H&E staining and specialized staining techniques for toxicology assessments and eluate models of autoimmune and metabolic diseases.

Figure A: H&E staining and various special staining methods for diverse animal models were used to evaluate adipose tissue morphology, colon structure, bone marrow integrity, inflammatory cell infiltration and fibrosis, lipid accumulation in the liver, epithelial cell layers in the intestine, and overall brain morphology.
